The bright spot near the center of the screen is a trapped bead, with a fluorecently stained DNA molecule attached. The flow cell holding the bead suspension is moved around in the plane of focus, making a loop in the DNA. The depth of focus of the trap, indicated by the short white bar near the top of the screen, is moved alternately up and down to thread the loop, thus making a trefoil knot.
