Abstract

This unit describes a ligation-mediated single-sided polymerase chain reaction (PCR) that is specifically designed for applications such as genomic footprinting and direct genomic sequencing. It should, however, be generally applicable to any PCR problem in which only one end of the region to be amplified is known.
