Experimental techniques play a central role in the development of molecular evolution. Imported mainly from molecular biology and biochemistry, different techniques have provided important new sources of data regarding molecular change.

From an experimental perspective, the history of molecular evolution can be divided into two periods: an earlier period where protein data dominated, and a later period beginning in the mid 1980s when DNA sequence data became more widely available.

A wide array of techniques were used in the development of molecular evolution, including microcompliment fixation, protein sequencing, hybridization, and DNA sequencing with PCR.
