[46] Factors influencing frequency production of transgenic mice

Abstract

[Introduction] Certain parameters have been defined that are essential for the efficient production of transgenic mice utilizing pronuclear injection of DNA. These include the concentration of DNA injected, composition of injection buffer, form of DNA, and mouse strain. Nevertheless, in the literature it is often revealed that good efficiency is not obtained. Therefore, in this discussion, we attempt to define some additional parameters that may influence success.

Each step involved in the production of transgenic mice is discussed with respect to what factors may influence efficiency. It is emphasized that not all comments are necessarily backed up by a wealth of experimental evidence. Some are based on limited observation by the authors.
